The reaction mixture contained in 1.0 ml: 3% azocasein, 100 μl of enzyme and 50 mM sodium acetate buffer 6.5 and adjusted to 1 ml with distilled water. The reaction mixture was preincubated at various temperatures for 15 min prior to substrate addition, followed by cooling in an ice bath.
