Comparison of mLysRS and mLysRS∆C for binding to IN and to tRNA₃^Lys. mLysRS and mLysRS∆C were expressed in E. coli with a C-terminal HA-tag. The HTRF assay, conducted as described in the legend of Fig. 4, was used to compare their binding constants to IN (a), and fluorescence polarization, conducted as described in the legend of Fig. 6, was used to compare their affinities for Cy-3 labeled tRNA₃^Lys (b).
