Altered processing of newly synthesized RGMc by PC inhibition. A. Time course of accumulation of RGMc on the cell surface and in conditioned medium after infection of Hep3B cells with Ad-HA-RGMc ± RVKR. Cell-surface proteins were labeled with non-permeable biotin (EZ-link), followed by incubation ± RVKR, and streptavidin pull-down, as described in 'Methods'. Immunoblot for cadherin measures sample loading. B. PC inhibition does not prevent acute release of RGMc from the cell surface. Membrane-associated RGMc was labeled with EZ-link, followed by incubation ± RVKR, and detection of soluble RGMc after streptavidin pull-down by immunoblotting.
