Time-course of binding of chitinase A and mutant enzymes to colloidal chitin. Chitinases (1 μmol in 100 mM sodium acetate buffer, pH 5.5) were incubated with 1.0 mg colloidal chitin at 0°C. Decreases in free enzyme concentration were determined at different time points from 0–30 min by Bradford's method. Each data value was calculated from triplicate experiments.
