IMP 5'-nucleotidase activity is undetectable in an isn1 mutant. Time course of hydrolysis of IMP (circles) or phenylphosphate (squares) in crude extracts from the isn1 knock-out strain (A) or wild-type strain (B). Reaction mixtures contained 10 mM IMP or phenylphosphate, 25 mM MgCl2, and 0.1% BSA in 100 mM imidazole/HCl (pH 6.5). The amounts of protein in the cell extracts used in these experiments were 270 μg for both isn1 knock-out and wild type strains.
