Specificity of the ARS1 binding. Gel retardation assays were performed in the absence (-) or presence (+) of GST-Cdc6 using radiolabeled ARS1 probe as described in Fig. 1B. The binding reaction was competed either with cold ARS1 fragment (lanes 3–6) or a 350 bp fragment from the plasmid BlueScript KS(+) digested with Hin fI followed by gel purification (lanes 7–10).
