Inhibition of AdsA 5'-nucleotidase activity by ADP. 2 μM rAdsA was incubated with 0-500 μM [14C]AMP in the presence of 75 μM cold AMP or ADP as indicated. (A) Radioactive signals for [14C]AMP and [14C]Ado after TLC were captured by a phosphorimager. Data is representative of two independent analyses. (B) Curve fitting analysis of data from (A) using nonlinear regression was performed using GraphPad Prism5 software. The results are expressed as nmols [14C]adenosine produced/min/mg rAdsA and are the averages of two experiments performed in duplicate. (C) Lineweaver-Burk plot of data in (B).
