Active site titration of wild-type EcMutY by binding to A/GO-containing DNA. DNA binding was assayed with 8 nM of A/GO-containing DNA and 2 to 200 nM (as measured by Bradford assay) of MutY. The result shows the average for two independent experiments. Calculated percentage bound were plotted as a function of EcMutY concentration and fitted by a modified version of Eq. 2 except [P]active is replaced with protein concentration determined by Bradford assay. The increase in wild-type (WT) EcMutY binding reached a maximum at a concentration about 20 nM of MutY and 5.44 nM DNA was bound (68% of 8 nM). This line (horizontal dotted line) intercepts with the slope line of initial rate (slant dotted line) at 8 nM of EcMutY. Thus, the WT EcMutY preparation is 68% active.
