Binding of acyl-CoAs to BnDGAT1(1–116)His6. Panels A and B: Binding of oleoyl-CoA or erucoyl-CoA to BnDGAT1(1–116)His6. Lipidex-1000 binding assays were conducted in duplicate in 10 mM potassium phosphate buffer, pH 7.4, at 30°C with [1-14C]acyl-CoAs and 0.2 μM protein. Incubations were set up without protein to correct for acyl-CoA that was not adsorbed by the Lipidex-1000. Data points represent mean values. Panels C and D: Scatchard plot [26] analysis of the binding of 18:1-CoA or 22:1-CoA to BnDGAT1(1–116)His6.
