Inhibition of 5-HT induced responses by Lerisetron. tsA 201 cells transfected with 5-HT_3ASR cDNA were clamped at a holding potential of -60 mV and perfused with 4 μM 5-HT (lower trace) or 4 μM 5-HT and 0.3 nM lerisetron (upper trace). Lerisetron reduced the peak 5-HT induced currents from 2.0 nA to 1.0 nA with no change in the response characteristics. An IC₅₀ value was determined by a combination of similar experiments and determined to be 0.4 ± 0.03 nM.
